چکیده

By TAPIO RYTOMAA P HLOXINE in propylene glycol provides a good and simple method of staining for the counting of eosinophilic granulocytes in the counting chamber. No comparable method has been available for the counting of neutrophils. The identification of cell types in certain excreta of the organism, especially in the intestinal contents, is a cumbersome procedure.”2 It has, however, proved necessary for quantitative studies of granulocyte kinetics.2’3 The present experimental investigation introduces a staining method based on the utilization of myeloperoxidase, an enzyme specific for the myeloid cell series.4-6 This method makes it possible to differentiate between granulocytes and other cells both in the counting chamber and in smears.

Role of hydrogen peroxide and peroxidase in the cytotoxicity of Trypanosoma dionisii by human granulocytes.

The mechanism of the cytotoxic reaction of leukocytes to Trypanosoma dionisii was investigated. Cytotoxicity was measured by release of [99mTc]pertechnetate from labeled protozoa. Both granulocytes and lymphocytes were found to be cytotoxic to antibody-coated T. dionisii. The reaction was inhibited by diethyldithiocarbamate and by potassium cyanide, both of which inhibit myeloperoxidase. Myelop...

Improving precision in the assessment of round cell numbers in human semen.

The aim of this study was to increase the precision of assessment of the number of round cells observed in the peroxidase test for detection of seminal leukocytes (granulocytes). The dilution of semen was reduced and the volume of suspension examined was increased for semen samples containing between 0.6 and 6 million round cells per mL. A 1 + 5 (1:6) dilution was compatible with measurable per...

Human granulocyte generation of hydroxyl radical

Human granulocytes were capable of oxidizing 2-keto-4 thiomethylbutyric acid to ethylene during phagocytosis or membrane perturbation. The reaction required hydrogen peroxide and superoxide and in addition was inhibited by various hydroxyl radical (OH) scavengers. These observations represent direct evidence for the generation of OH by human granulocytes. Further, inhibition of ethylene generat...

Use of rapid cytochemical staining to characterize fish blood granulocytes in species of special concern and determine potential for function testing.

Studies of innate immunity in fish species of special concern are essential for better understanding of their health status during hatchery rearing conditions. The cytochemical and morphological characterizations of blood granulocytes have been used to provide information about phylogenetic differences and determine the potential use of neutrophil function assays. Rapid, simple, cytochemical st...

Bisubstrate Kinetic Model for Enzymatic Decolorization of Reactive Black 5 by Coprinus cinereus Peroxidase

In this study, decolorization of the diazo dye, Reactive Black 5 (RB5) in a Coprinus cinereus peroxidase-catalyzed reaction has been investigated. A bisubstate kinetic model for the reaction rate based on the Ping-Pong mechanism was assumed for the enzymatic decolorization. Experiment...
